AbstractThe research aims to increase the utilization of re sidue from corn milk production that mostly used for feeding stock. Zein is a main protein in t he corn kernel and widely used for film and coating. Therefore, in the present study, zein was extract ed from corn milk residue using aqueous ethanol. The fresh corn milk residue received from Suwanwajckka sikit Field Corps Research Station (Thailand) was dried by tray dryer to obtain 8-10 percent mo isture content prior to extraction. The drying temperatures were 70, 90 and 120 o C. The yields of crude zein prot eins were studied. The result showed that yields of zein obtained fr om both residues that dried at 70 and 90 o C were not significant different and greater than that of residue from 120 o C. An effect of extr action temperature (25-80 o C) on the extraction yield was further studied. The result showed yields and proteins content of crude zein obtained from those extracted at 60 and 80 o C were significant greater than those extracted at 25 and 40 o C. An effect of NaOH was also elucidated. The result showed yields of crude zein obtained from the conditions such as with and without NaOH, 0.05 and 0.08 N were not significant different. However, protein content of crude zein ex tracted using 70 percent ethanol adding 0.05 N NaOH was greater than that extracted without NaOH. The ge l electrophoresis (4% stacking gel and 12.5 % separating gel) determined the subunit of extract ed zein compared to the commercial zein was done. SDS-polyacrylamide gel patterns of the extracted zein appeared to be similar to that of commercial zein. ? -Zein (22-24 kDa) was found to be the major protein of the extracted zein.
